Is it possible to create 45 columns with the same settings at one time?

Sure. Just a couple of steps, from the gradebook view:

1- Add column > Favorite > Add favorite and
configure here the default settings for those columns.

2- Add column > Multiple columns > Favorite
Select here the favorite column configuration you've just created
and type 45 in the number of columns (if you tap on the center a numeric keypad will appear)
